我室将MTT法用于检测短小棒状扦菌(CP)对小鼠腹腔Mφ的激活效应,获得较满意结果.1 材料和方法1.1 材料 短小棒状杆菌(CorynebacteriumParvum,CP,H-84),.河南医科大学微生物学教研室惠赠,每毫昇含福尔马林灭活菌苗2mg.1.2 CP使用方法 选NIH小鼠,雌性,体重20g±2g.腹腔注射CP50mg/kg体重(约0.5ml/只),对照组注射等体积生理盐水.注射后不同时间分别杀鼠,常规制备腹腔Mφ.1.3 腹腔Mφ功能测定参照文献[1~4] 采用MTT法,测定A值,检测波长570nm,参考波长630nm.
MTT assay was used to detect the activation of Mφ in mouse peritoneal cavity by MTT method.Materials and Methods1.1 Materials Corynebacterium Parvum (CP, H-84), Henan Medical University Department of Microbiology, gift per milliliter containing formalin inactivated vaccine 2mg.1.2 CP use NIH mice, female, weighing 20g ± 2g. Intraperitoneal injection of CP50mg / kg body weight (about 0.5ml / only) , The control group were injected with equal volume of normal saline.After injection, mice were sacrificed at different times and routinely prepared for intraperitoneal Mφ.1.3 intraperitoneal Mφ function determination [1 ~ 4] MTT method was used to determine the value of A, the detection wavelength of 570nm, the reference wavelength of 630nm.
