目的 探讨胰腺癌组织端粒酶活性和端粒酶基因表达及两者的相关性 ,及其与癌生物学行为的关系 ,以评价端粒酶活性和端粒酶基因表达检测对胰腺癌的诊断价值。方法 采用TRAP -PCR -ELISA方法和原位杂交方法 ,检测了 2 3例胰腺癌患者端粒酶活性和基因表达情况。结果 2 3例胰腺癌组织端粒酶阳性率为 87.0 % ,10例癌周组织阳性率为 10 .0 % ,两者有显著性差异 (P <0 .0 5 )。 2 3例胰腺癌组织中 19例 (82 .6% )hTR表达阳性 ,10例癌周组织中 1例 (10 .0 % )hTR表达阳性。胰腺癌组织中hTR表达阳性率与癌周组织比较 ,有显著性差异 (P <0 .0 5 )。端粒酶活性及其基因表达与胰腺癌组织学分级、临床分期及伴随局部淋巴结转移无明显相关性 (P >0 .0 5 )。结果还显示端粒酶活性与hTR基因表达相关 (P <0 .0 5 )。结论 端粒酶可能是胰腺癌发生的早期事件 ,端粒酶活性及端粒酶基因表达的检测对胰腺癌具有一定的诊断价值
Objective To investigate the correlation between telomerase activity and telomerase gene expression in pancreatic cancer and its relationship with the biological behavior of pancreatic cancer in order to evaluate the diagnostic value of telomerase activity and telomerase gene expression in the diagnosis of pancreatic cancer value. Methods TRAP-PCR-ELISA and in situ hybridization were used to detect telomerase activity and gene expression in 23 patients with pancreatic cancer. Results The positive rate of telomerase in 23 cases of pancreatic cancer was 87.0%, and the positive rate of 10 cases was 10.0%. There was a significant difference between the two (P <0.05). HTR expression was positive in 19 (82.6%) of the 23 pancreatic cancer tissues and 1 (10.0%) in 10 of the peritumoral tissues. The positive rate of hTR expression in pancreatic cancer tissues compared with the peritumoral tissue, there was a significant difference (P <0.05). There was no significant correlation between telomerase activity and gene expression and pancreatic cancer histological grade, clinical stage and accompanying lymph node metastasis (P> 0.05). The results also showed that telomerase activity was associated with hTR gene expression (P <0.05). Conclusion Telomerase may be an early event in pancreatic cancer. The detection of telomerase activity and telomerase gene expression has certain diagnostic value for pancreatic cancer
