目的:观察IL1的受体拮抗剂(IL1ra)对缺氧性神经细胞的影响,探讨IL1在神经细胞缺氧损伤中的作用以及基因转染治疗的可能性。方法:(1)以缺陷型HSV为载体构建的pHSVIL1ra转染体外培养的神经细胞为实验组,以pHSVLacZ转染的体外培养的神经细胞为对照,进行体外缺氧实验。以神经细胞的3HGABA和3HGlu摄取能力及细胞凋亡情况为判定损伤程度的指标。结果:转染pHSVIL1ra的神经细胞缺氧后,其3HGABA和3HGlu的摄取较转染pHSVLacZ的神经细胞有所增高,神经细胞凋亡的程度少于对照。结论:pHSVIL1ra转染可减轻缺氧引起的神经细胞急性损伤和迟发性损伤。
Objective: To observe the effect of IL1 receptor antagonist (IL1ra) on hypoxic neurons and to explore the role of IL1 in hypoxic injury of nerve cells and the possibility of gene transfection therapy. Methods: (1) In vitro culture of nerve cells transfected with pHSVIL1ra constructed by defective HSV vector was used as experimental group. In vitro hypoxia experiments were carried out in vitro using pHSVLacZ transfected neural cells as control. 3H-GABA neurons and 3H Glu uptake capacity and apoptosis as indicators of the degree of injury. Results: After hypoxia transfected neurons in pHSVIL1ra cells, the uptake of 3HGABA and 3HGlu in neurons was higher than that in pHSVLacZ transfected cells, and the degree of neuronal apoptosis was less than that in the control. CONCLUSIONS: Transfection with pHSVIL1ra can reduce acute and delayed neuronal injury caused by hypoxia.
