目的:研究温州地区胃癌与瘦素受体Pro1019 Pro基因变异的关系。方法:运用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法测定无亲缘关系的221例温州地区汉人的瘦素受体基因Pro1019 Pro的基因型(其中胃癌组117例,正常对照组104例),并分析Pro1019 Pro基因多态性与糖尿病的关系。结果:①胃癌组Pro1019 Pro基因型GG、GA及AA的频率分别为0.017、0.205和0.778;G和A等位基因频率分别为0.120和0.880;正常对照组基因型GG、GA及AA的频率分别为0.077、0.365和0.558;G和A等位基因频率分别为0.260和0.740;②胃癌组A、G等位基因分布频率与正常对照组比较差异有显著性(P<0.05)。结论:温州地区汉族人群存在瘦素受体基因Pro1019 Pro变异,它可能是该地区人群胃癌患者的遗传易感标记,A等位基因与胃癌发病有一定关系。
Objective: To study the relationship between gastric cancer and leptin receptor Pro1019 Pro mutation in Wenzhou area. Methods: The genotypes of 221 unrelated Chinese Han Chinese leptin receptor gene Pro1019 Pro were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) (117 in gastric cancer group, normal Control group of 104 cases), and analyze the relationship between Pro1019 Pro gene polymorphism and diabetes. Results: ① The frequencies of GG, GA and AA of Pro1019 Pro genotype in gastric cancer group were 0.017,0.205 and 0.778 respectively; the allele frequencies of G and A alleles were 0.120 and 0.880 respectively; the frequencies of GG, GA and AA genotype in normal control group were 0.077,0.365 and 0.558, respectively. The frequencies of G and A alleles were 0.260 and 0.740, respectively. The frequencies of A and G alleles in gastric cancer group were significantly different from those in normal controls (P <0.05). Conclusion: Pro1019 Pro mutation of leptin receptor gene exists in Wenzhou Han population. It may be a genetic susceptible marker for gastric cancer patients in this area. A allele is associated with the development of gastric cancer.
