目的:通过对老化的紫苏种子萌发及幼苗生理特性的研究,寻找提高紫苏种子及幼苗抗老化能力的途径。方法:测定不同浓度硫酸锌(ZnSO4)和聚乙二醇(PEG)处理后的紫苏种子的发芽势、发芽率、发芽指数和活力指数,并对紫苏幼苗叶片的丙二醛(MDA)含量、超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性进行测定。结果:老化的紫苏种子在经过不同浓度的ZnSO4和PEG处理后,萌发指标均有升高,其中经过600 mg.L-1的ZnSO4和20%的PEG处理后,各项指标均达到最大值,发芽势(Gv)分别为64.7,66.8%;发芽率(Gr)分别为78.7%,79.4%;发芽指数(Gi)分别为11.8,12.2;活力指数(Vi)分别为0.091 1,0.093 9。不同处理均降低了叶片MDA的含量,并且提高了SOD,POD,CAT3种酶的活性,显著的提高了幼苗生物量的积累。当ZnSO4为600 mg.L-1时,3种酶活性分别为0.28,4.71,3.82 U.mg-1,当PEG浓度为20%时,3种酶的活性分别为0.29,4.93,4.18 U.mg-1。结论:600 mg.L-1的ZnSO4和20%的PEG处理能够有效的减缓老化对紫苏种子及幼苗产生的伤害,提高了种子及幼苗的抗老化能力。
OBJECTIVE: To find a way to improve the anti-aging ability of Perilla frutescens seeds and seedlings by studying the germination and the physiological characteristics of the aged Perilla frutescens seeds. Methods: The germination energy, germination rate, germination index and vigor index of Perilla frutescens seeds treated with different concentrations of ZnSO4 and PEG were measured. Malondialdehyde (MDA) Content, superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activity were measured. Results: The germination indexes of aged Perilla frutescens seeds increased after treated with different concentrations of ZnSO4 and PEG. After the treatment with 600 mg.L-1 ZnSO4 and 20% PEG, the indexes all reached the maximum , Germination potential (Gv) were 64.7,66.8% respectively; germination rate (Gr) were 78.7% and 79.4% respectively; germination index (Gi) was 11.8 and 12.2, and vigor index was 0.091 and 1.093 respectively. All the treatments decreased the content of MDA and increased the activities of SOD, POD and CAT3 enzymes, and significantly increased the biomass accumulation of seedlings. When ZnSO4 was 600 mg.L-1, the activities of the three enzymes were 0.28, 4.71 and 3.82 U.mg-1 respectively. When the concentration of PEG was 20%, the activities of the three enzymes were 0.29, 4.93 and 4.18 U, respectively. mg-1. CONCLUSION: The treatment of 600 mg.L-1 ZnSO4 and 20% PEG can effectively slow down the damage caused by aging to perilla seeds and seedlings, and improve the anti-aging ability of seeds and seedlings.
