目的探讨骨形成发生蛋白4(BMP4)和SMAD4表达变化在胶质瘤诊断中的意义。方法随机选取胶质瘤标本45例,正常脑组织10例,应用RT-PCR和Western blot检测胶质瘤标本和正常脑组织中BMP4和SMAD4表达。结果 RT-PCR结果显示,与正常脑组织相比,低级别胶质瘤、高级别胶质瘤中BMP4表达分别降低(23.5±0.23)%、(46.6±0.34)%,差异有统计学意义(P<0.05)。SMAD4表达分别降低(30.6±0.41)%、(95.3±1.02)%(P<0.05)。Western blot结果显示与正常脑组织相比,低级别胶质瘤、高级别胶质瘤BMP4表达分别降低(28.4±0.25)%、(80.4±1.05)%(P<0.05)。SMAD4表达降低(76.8±1.15)%、(96.2±2.01)%(P<0.05),差异有统计学意义。结论 BMP4、SMAD4可能与胶质瘤的发生发展及恶性程度相关,BMP4、SMAD4可能成为胶质瘤诊断和分级的一个重要指标。
Objective To investigate the significance of the expression changes of bone morphogenetic protein 4 (BMP4) and SMAD4 in the diagnosis of gliomas. Methods Forty-five glioma samples and 10 normal brain samples were randomly selected. The expressions of BMP4 and SMAD4 in glioma specimens and normal brain tissues were detected by RT-PCR and Western blot. Results Compared with normal brain tissue, the expression of BMP4 in low grade gliomas and high grade gliomas was significantly lower (23.5 ± 0.23)% and (46.6 ± 0.34)%, respectively, compared with normal brain tissue P <0.05). SMAD4 expression decreased by (30.6 ± 0.41)% and (95.3 ± 1.02)%, respectively (P <0.05). Western blot results showed that compared with normal brain tissue, the expression of BMP4 in low grade gliomas and high grade gliomas decreased by (28.4 ± 0.25)% and (80.4 ± 1.05)%, respectively (P <0.05). SMAD4 expression decreased (76.8 ± 1.15)%, (96.2 ± 2.01)% (P <0.05), the difference was statistically significant. Conclusion BMP4 and SMAD4 may be related to the development and malignancy of glioma. BMP4 and SMAD4 may be an important index for the diagnosis and grading of gliomas.
