在抗心律失常、抗心绞痛及抗高血压治疗中,常常联合应用钙拮抗剂和强心甙。大量研究工作已显示,同时服用维拉帕米及狄戈辛后,血浆狄戈辛浓度显著升高达60%到70%,从而增加了狄戈辛过量的危险。给予维拉帕米后,肾脏的狄戈辛清除减少被认为是血浆狄戈辛浓度增高的主要机制。与狄戈辛相比,洋地黄毒甙(digitoxin)的排泄不受肾功能的影响。本文旨在估价治疗剂量的维拉帕米对洋地黄毒甙在前瞻性临床模武中的药代动力学及心脏效应的影响。
In the anti-arrhythmia, anti-angina and antihypertensive treatment, often combined with calcium antagonist and cardiac glycosides. Numerous studies have shown that with the simultaneous administration of verapamil and Digoxiin, there is a marked increase in the concentration of digoxin from 60% to 70%, thereby increasing the risk of excessive digoxin. After the administration of verapamil, the reduction of digoxigenin in the kidneys is considered to be the major mechanism of increased plasma digoxin concentration. Compared with Digoxin, digitoxin excretion is not affected by renal function. This article aims to evaluate the effect of verapamil at a therapeutic dose on the pharmacokinetics and cardiac effects of digitoxin in prospective clinical modalities.
