目的 探讨GM CSF联合IL 4对髓性白血病细胞表面B7分子表达的上调作用。方法 从初诊急性或慢性髓性白血病患者的外周血中分离出单个核细胞 (PBMNC) ,用rhGM CSF联合rhIL 4共同孵育 7~ 10d ;通过流式细胞仪检测细胞因子诱导前后细胞表面B7分子的表达。结果 未经细胞因子诱导的白血病细胞表面B7分子低表达或缺如 ,经过GM CSF联合IL 47~ 10d的诱导 ,可显著上调B7分子的表达。结论 髓性白血病细胞表面存在B7分子表达缺陷 ;用GM CSF联合IL 4进行诱导是纠正这种缺陷的一条途径
Objective To investigate the effect of GM CSF combined with IL 4 on the upregulation of B7 expression on myeloid leukemia cells. Methods Mononuclear cells (PBMNC) were isolated from the peripheral blood of patients with newly diagnosed acute or chronic myeloid leukemia and incubated with rhGM CSF and rhIL 4 for 7 to 10 days. Flow cytometry was used to detect B7 molecules on the cell surface before and after cytokine induction. expression. Results Without the low expression or absence of B7 molecules on the surface of leukemia cells induced by cytokines, the expression of B7 molecules could be significantly up-regulated after induction of GM CSF combined with IL 47-10 days. Conclusion B7 molecule expression defects exist on the surface of myeloid leukemia cells; induction with GM CSF combined with IL 4 is a way to correct this defect.
