目的:研究补肾壮骨中药(主要由淫羊藿、续断、补骨脂等组成)对糖皮质激素诱发的骨质疏松大鼠骨代谢、骨生物力学性能的影响。方法:30只3.5月龄雌性SD大鼠随机分为3组,每组10只,即正常对照组、模型组、补肾壮骨中药组。皮下注射甲泼尼龙(3.5 mg/kg,9 w),建立骨质疏松模型。补肾壮骨中药组造模后给予仙灵骨葆胶囊(150 mg/kg)灌胃,2/d,其他2组给予等量生理盐水,实验期9 w。9 w后测定各组大鼠骨密度(BMD)、骨代谢生化指标及骨生物力学指标等。结果:与对照组比较,模型组大鼠BMD、骨生物力学指标、血清Ca、BGP、CT水平均显著降低(P<0.05),TRACP酶活性与血清P水平无显著性变化(P>0.05);与模型组比较,补肾壮骨中药组大鼠BMD、血清Ca、BGP、CT水平均显著升高(P<0.05),但对TRACP酶活性、血清P水平无显著性影响(P>0.05)。结论:补肾壮骨中药可通过改善骨代谢生化指标及骨生物力学性能,拮抗糖皮质激素性骨质疏松大鼠的骨丢失,起到防治骨质疏松的作用。
Objective: To study the effect of Bushen Zhuanggu (mainly composed of Epimedium, Dipsacus, Psoralen) on bone metabolism and bone biomechanical properties in rats with glucocorticoid-induced osteoporosis. Methods: Thirty female 3.5-month-old female Sprague-Dawley rats were randomly divided into three groups (n = 10 each): normal control group, model group and Bushen Zhuanggu Chinese Herbal Medicine group. Methylprednisolone (3.5 mg / kg, 9 w) was injected subcutaneously to establish an osteoporosis model. Bushen Zhuanggu was given Xianling Gubao capsule (2mg / kg) for 2 days after modeling, and the other two groups were given the same amount of normal saline for 9 w. After 9 weeks, the bone mineral density (BMD), biochemical markers of bone metabolism and biomechanical parameters of bone were measured. Results: Compared with the control group, BMD, bone biomechanical parameters, serum Ca, BGP and CT levels in model group were significantly decreased (P <0.05), TRACP enzyme activity and serum P level had no significant change (P> 0.05) Compared with the model group, the levels of BMD, serum Ca, BGP and CT in rats of Bushenzhuanggao group were significantly increased (P <0.05), but had no significant effect on TRACPase activity and serum P level (P> 0.05) . Conclusion: Bushen Zhuanggu Chinese herbs can prevent osteoporosis by improving the biochemical indexes of bone metabolism and biomechanical properties of bone to antagonize the bone loss of glucocorticoid-induced osteoporosis rats.
