PSD-007单体是已确定结构式的血卟啉类激光光敏剂。我们用人大肠癌细胞株SW1116,宫颈癌细胞株HeLa和红白血病细胞株K562,以北京产的同类光敏剂HPD作为药物对照,在波长630nm的铜蒸汽激光照射4J/cm2能量密度后,用MTT法测定两种光敏剂对3个人癌细胞的9个浓度组,计算50%细胞抑制率(IC50)和细胞生存率的均数,比较两个光敏剂对三株人癌细胞的光动力杀伤效应。实验结果显示:用K562细胞HPD的IC50为4mg/L,PSD-007为4mg/L;HeLa细胞IC50为4.5mg/L和4mg/L;SW1116细胞的IC50为4.5mg/L和1mg/L。在对两种药物7个不同浓度研究组中,抑瘤结果表明,各组间有显著性差异(P<0.05),说明随着药物浓度的增高光动力杀伤效应增大。
PSD-007 monomer is a hematoporphyrin-based laser photosensitizer with a defined structure. We used human colorectal cancer cell line SW1116, cervical cancer cell line HeLa, and erythroleukemia cell line K562, and used Beijing-based photosensitizer HPD as a drug control. After irradiation with a copper vapor laser at a wavelength of 630 nm and an energy density of 4 J/cm2, the MTT method was used. Two photosensitizers were used to measure 9 concentrations of 3 human cancer cells. The mean 50% cell inhibition rate (IC50) and cell survival rate were calculated. Photodynamic killing effects of two photosensitizers on three human cancer cells were compared. The experimental results showed that the IC50 of HPD in K562 cells was 4 mg/L, PSD-007 was 4 mg/L; the IC50 of HeLa cells was 4.5 mg/L and 4 mg/L; the IC50 of SW1116 cells was 4.5 mg/L and 1 mg/L. L. In the seven different concentrations of the two drugs in the study group, tumor inhibition results showed that there was a significant difference between the groups (P <0.05), indicating that with the increase of drug concentration, photodynamic killing effect increased.
