目的:研究糖泰胶囊对实验性2型糖尿病(T2DM)大鼠血糖、血脂代谢的影响。方法:采用高脂饮食联合腹腔注射链脲佐菌素(STZ 30 mg·kg-1)诱导制备T2DM大鼠模型,将造模成功大鼠随机分为模型组、二甲双胍组、糖泰胶囊高、中、低(8.64 g·kg-1、4.32 g·kg-1、2.16 g·kg-1)剂量组,每组10只,取10只正常大鼠为正常对照组,各组予相应的药物干预4周。生化法检测大鼠空腹血糖、口服葡萄糖耐量、糖化血红蛋白、血脂水平;放射免疫法检测血清胰岛素水平。结果:模型组大鼠口服糖耐量异常,血糖值、糖化血红蛋白、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)显著高于正常对照组,高密度脂蛋白胆固醇(HDL-C)、胰岛素水平低于正常对照组,差异均有统计学意义(P<0.01或P<0.05);干预4周后,二甲双胍组、糖泰胶囊高、中剂量组大鼠空腹血糖均显著下降(P<0.01或P<0.05)。糖泰胶囊高剂量组糖负荷后0.5 h血糖和血糖曲线下面积(AUC)均小于模型组(P<0.05)。糖泰胶囊高、中剂量组Hb A1c水平低于模型组,胰岛素水平高于模型组,差异均有统计学意义(P<0.01或P<0.05)。糖泰胶囊高剂量组大鼠血清TC,TG、LDL-C均低于模型组,HDL-C高于模型组,但差异均无统计学意义(P>0.05)。结论:糖泰胶囊能降低2型糖尿病大鼠血糖值、糖化血红蛋白,升高血清胰岛素水平,改善其口服糖耐量,但对血脂调节无明显影响。
Objective: To investigate the effect of Tangtai capsule on blood glucose and lipid metabolism in experimental type 2 diabetes mellitus (T2DM) rats. Methods: T2DM rats were induced by high-fat diet combined with streptozotocin (STZ 30 mg · kg-1). The rats were randomly divided into model group, metformin group, Medium and low (8.64 g · kg -1, 4.32 g · kg -1 and 2.16 g · kg -1) groups, 10 rats in each group. Ten normal rats were selected as the normal control group, and the corresponding drugs Intervention 4 weeks. The fasting blood glucose, oral glucose tolerance, glycosylated hemoglobin and blood lipids in rats were detected by biochemical methods. Serum insulin levels were measured by radioimmunoassay. Results: The oral glucose tolerance, blood glucose, glycosylated hemoglobin, total cholesterol (TC), triglyceride (TG) and low density lipoprotein cholesterol (LDL-C) in the model group were significantly higher than those in the normal control group (P <0.01 or P <0.05). After intervention for 4 weeks, the rats in metformin group and high-dose group of Tangtai Capsule had higher levels of serum HDL-C and insulin than the normal control group Fasting plasma glucose were significantly decreased (P <0.01 or P <0.05). The area under the curve of blood sugar and blood glucose 0.5 h after high-dose Tangtai capsule treatment was lower than that of model group (P <0.05). The levels of Hb A1c in high dose and middle dose groups of Tangtai Capsule were lower than those in model group, while the insulin level was higher in model group than in model group. The difference was statistically significant (P <0.01 or P <0.05). The levels of TC, TG and LDL-C in high-dose Tangtai capsule group were significantly lower than those in model group, and HDL-C was higher in model group than in model group (P> 0.05). Conclusion: Tangtai Capsule can reduce blood glucose, HbA1c, increase serum insulin and improve oral glucose tolerance in type 2 diabetic rats, but has no significant effect on blood lipid regulation.
