对PEG(聚乙二醇)/Mg SO4双水相萃取苦苣皂甙进行研究,比较双水相萃取的皂甙与其它常用提取方法提取的皂甙的抗氧化性。选取PEG4000浓度、Mg SO4浓度、p H、温度为独立变量,苦苣皂甙回收率为指标,采用Box-benhnken响应面法优化,得到回归模型。结果表明:各变量的影响顺序为PEG4000浓度>p H>萃取温度>Mg SO4浓度;由回归分析得到最佳工艺条件是:PEG4000质量分数21%,Mg SO4质量分数16%,萃取温度51℃,p H 6,此时苦苣皂甙回收率为95.12%。双水相萃取所得皂甙与其它方法所得皂甙的提取率相差不大。苦苣皂甙抗氧化活性强弱与皂甙浓度呈量效关系,且与提取方法有关;双水相萃取所得皂甙抗氧化活性最强。
The extraction of chicory saponin from PEG (polyethylene glycol) / MgSO 4 aqueous two-phase was studied, and the antioxidant properties of saponins extracted from aqueous two-phase extraction and other commonly used extraction methods were compared. The PEG4000 concentration, MgSO4 concentration, pH, temperature as independent variables, the recovery rate of chicory saponin as an indicator, using Box-benhnken response surface method optimization, regression model was obtained. The results showed that the order of the variables was PEG4000 concentration> p H> extraction temperature> MgSO4 concentration. The optimum technological conditions obtained by regression analysis were as follows: PEG4000 content 21%, MgSO4 content 16%, extraction temperature 51 ℃, p H 6, then the recovery rate of chicory saponin was 95.12%. The extraction yield of saponins from aqueous two-phase extraction was similar to other methods. The antioxidant activity of chicory saponin and saponin concentration showed a dose-effect relationship, and the extraction method; saponin obtained by aqueous two-phase extraction strongest antioxidant activity.
