目的:观察浓缩归脾丸对苯中毒并抑郁小鼠的定时游泳总里程、行为学及缺氧存活时间等应激能力的影响。方法:将72只小鼠随机分成6组,即正常组、模型组、归脾丸高、中、低各剂量组和西药组,每组12只。除正常组外,其余组均采用熏苯14天,并随机刺激14天建立苯中毒并应激抑郁小鼠模型,在造模成功后第1天开始给药。正常组、模型组均给予生理盐水0.2 mL/只灌胃;归脾丸高、中、低剂量组分别按8、4、2 mg/(kg·d)灌胃;西药组给予利血生1.5 mg/(kg·d),鲨肝醇5 mg/(kg·d),配制成混悬液灌胃;每天1次,连续3周。检测小鼠3 min定时游泳总里程、行为学及缺氧存活时间等指标。结果:与模型组比较,正常组3 min定时游泳总里程和小鼠的行为得分显著较高,缺氧存活时间显著较长,差异均有统计学意义(P<0.05,P<0.01);归脾丸各剂量组及西药组3 min定时游泳总里程显著延长,归脾丸各剂量组的垂直得分和水平得分明显升高,归脾丸各剂量组及西药组均能延长苯中毒小鼠的缺氧存活时间,差异均有统计学意义(P<0.05,P<0.01);与西药组比较,归脾丸各剂量组3 min定时游泳总里程、垂直和水平得分升高的更明显,归脾丸中、低剂量组小鼠的缺氧存活时间更长,差异均有统计学意义(P<0.05)。结论:归脾丸可提高苯中毒并抑郁小鼠的应激能力并减轻其抑郁的程度。
Objective: To observe the effects of concentrated Guipi Pill on total swimming time, behavior and hypoxic survival time of benzene poisoning and depression mice. Methods: 72 mice were randomly divided into 6 groups, namely normal group, model group, Guipiwan high, medium and low dose groups and western medicine group, 12 rats in each group. In addition to the normal group, the remaining groups were smoked benzene 14 days, and 14 days of random stimulation to establish benzene poisoning and stress depression in mice model, the first day after the success of the model administration. Results: Compared with the model group, the total swimming time and the behavioral score of the mice in the normal group at 3 min were significantly higher and the hypoxia survival time was significantly longer (P <0.05, P <0.01). Spleen pill dose groups and Western medicine group 3 min total swimming time significantly extended, Guipiwan dose group of the vertical score and the level of scores was significantly increased, Guipiwan dose group and western medicine group can extend the benzene poisoning mice (P <0.05, P <0.01). Compared with Western medicine group, the total swimming time, vertical and horizontal swimming time of each dose group of Guipiwan increased more obviously In spleen pill, low dose group of mice hypoxia survival longer, the difference was statistically significant (P <0.05). Conclusion: Guipi Pill can increase benzene stress and depress the stress ability of mice and reduce the degree of depression.
