目的探讨下调婆罗双树样基因4(SALL4)对肾癌ACHN细胞周期和细胞凋亡的影响。方法根据人SALL4的mRNA序列设计合成靶向SALL4的小干扰RNA(siRNA),用于沉默ACHN细胞SALL4的表达。运用实时定量PCR(qRT-PCR)和Western blot法分别检测SALL4的mRNA与蛋白表达水平;使用流式细胞术检测ACHN细胞的细胞周期和细胞凋亡情况。结果 SALL4 siRNA可显著下调ACHN细胞中SALL4的mRNA和蛋白表达水平,下调SALL4后ACHN细胞的细胞周期受到抑制,G1期细胞增多,S/G2期减少,细胞凋亡显著增加。结论下调SALL4基因表达可抑制肾癌ACHN细胞增殖并促进其凋亡,SALL4可能是肾癌治疗的新靶点。
Objective To investigate the effects of SALL4 on the cell cycle and apoptosis of ACHN cells in renal cell carcinoma. Methods A small interfering RNA (siRNA) targeted to SALL4 was designed and synthesized according to human SALL4 mRNA sequence, which was used to silence the expression of SALL4 in ACHN cells. The mRNA and protein expression of SALL4 were detected by real-time quantitative PCR (qRT-PCR) and Western blot respectively. The cell cycle and apoptosis of ACHN cells were detected by flow cytometry. Results SALL4 siRNA significantly down-regulated the mRNA and protein expression of SALL4 in ACHN cells. After SALL4 down-regulation, the cell cycle of ACHN cells was inhibited, S phase G1 cells increased, S / G2 phase decreased, and apoptosis increased significantly. Conclusion Down-regulation of SALL4 gene expression can inhibit the proliferation and promote the apoptosis of ACHN cells in renal cell carcinoma. SALL4 may be a new target for the treatment of renal cell carcinoma.
