目的 研究亚低温对脑创伤后谷氨酸细胞外释放及c fosmRNA表达的影响。方法 原位杂交法并图像分析系统来评估c fosmRNA表达。以脑脊液的谷氨酸含量评价其细胞外的释放程度。随机分为亚低温和常温组。每一组又分假手术和伤后 10、30、6 0、12 0分钟亚组。结果 常温组谷氨酸明显高于亚低温组 (P <0 .0 5 ) ,而c fosmRNA产物在亚低温组明显高于常温组 (P <0 .0 5 ) ,假手术组无阳性细胞。结论 亚低温能显著降低创伤性脑损伤引起的谷氨酸细胞外释放 ,同时更能促进c fosmRNA表达。
Objective To investigate the effects of mild hypothermia on extracellular glutamate release and c fos mRNA expression after traumatic brain injury. Methods In situ hybridization and image analysis systems were used to assess c fos mRNA expression. The degree of extracellular release of cerebrospinal fluid was evaluated by its glutamate content. Randomly divided into mild hypothermia and normal temperature group. Each group was divided into sham operation and 10,30,6 0,12 minutes after injury group. Results The level of glutamate in normal temperature group was significantly higher than that in mild hypothermia group (P <0.05), while c fos mRNA was significantly higher in hypothermia group than in normal temperature group (P <0.05). There was no positive cells in sham operation group. Conclusion Mild hypothermia can significantly reduce the release of extracellular glutamate induced by traumatic brain injury and promote the expression of c fos mRNA.
