目的探讨中电导钙激活钾离子通道(Intermediate-conductance calcium-activated potassium channel,SK4)在乳腺癌组织和细胞系中的表达及其临床意义。方法 42例乳腺癌组织标本,20例距肿瘤切缘2 cm的癌旁组织标本。免疫组织化学法(Immunohistochemistry,IHC)和Western Blot法(WB)检测SK4在乳腺癌组织和乳腺癌细胞系中的表达,并分析其与临床病理指标的关系。结果 SK4在癌组织中的表达高于癌旁组织。SK4在乳腺癌组织和癌旁组织中的表达阳性率分别为88.1%和35.0%(P<0.05);低、中、高分化肿瘤中SK4表达阳性率分别为100.0%、87.5%和75.0%(P<0.05);伴或不伴淋巴结转移的患者SK4的表达阳性率分别为69.2%和99.6%(P<0.05);在乳腺癌雌激素受体阳性和阴性中SK4的阳性率别为86.7%和91.7%(P>0.05)。SK4在乳腺癌细胞系MDA-MB-231和MCF-7中明显表达。结论 SK4在乳腺癌组织中表达增加;SK4的表达与肿瘤分化程度、淋巴结转移相关。
Objective To investigate the expression of intermediate-conductance calcium-activated potassium channel (SK4) in breast cancer tissues and cell lines and its clinical significance. Methods 42 cases of breast cancer tissue specimens, 20 cases from the tumor resection margin of 2 cm paraneoplastic tissue specimens. Immunohistochemistry (IHC) and Western Blot (WB) were used to detect the expression of SK4 in breast cancer and breast cancer cell lines, and analyzed its relationship with clinicopathological parameters. Results The expression of SK4 in cancer tissues was higher than that in paracancerous tissues. The positive rates of SK4 expression in breast cancer tissues and paracancerous tissues were 88.1% and 35.0%, respectively (P <0.05). The positive rates of SK4 in low, moderate and well differentiated tumors were 100.0%, 87.5% and 75.0% P <0.05). The positive rates of SK4 expression in patients with and without lymph node metastasis were 69.2% and 99.6%, respectively (P <0.05). The positive rates of SK4 in estrogen receptor positive and negative breast cancer tissues were 86.7% And 91.7% (P> 0.05). SK4 was significantly expressed in breast cancer cell lines MDA-MB-231 and MCF-7. Conclusion The expression of SK4 is increased in breast cancer tissues. The expression of SK4 is correlated with the degree of tumor differentiation and lymph node metastasis.
