目的探讨骨髓间充质干细胞(BMSCs)在大鼠颌下腺细胞放射性损伤中的修复作用。方法采用60钴照射,建立大鼠颌下腺细胞损伤模型,将BMSCs与颌下腺细胞共培养。激光共聚焦显微镜下观察细胞形态,采用MTS法检测细胞增殖情况。结果 60钴照射5min可对体外培养的颌下腺细胞造成实验所需的放射性损伤。共培养细胞中部分BMSCs呈现颌下腺腺泡细胞形态,共培养细胞的增殖率高于经过放射性损伤的颌下腺细胞(P<0.01),而与正常颌下腺细胞和BMSCs相比无统计学差异(P>0.05)。结论 BMSCs具有转化为颌下腺腺泡细胞的潜力,对放射性涎腺细胞损伤起到一定的修复作用。
Objective To investigate the repair effect of bone marrow mesenchymal stem cells (BMSCs) on radiation-induced injury of submandibular gland cells in rats. Methods 60Co irradiation was used to establish rat submandibular gland cell injury model. BMSCs were co-cultured with submandibular gland cells. Cell morphology was observed under confocal laser scanning microscope. Cell proliferation was detected by MTS assay. Results 60Co irradiation for 5 min caused the experimental radiation damage to the cultured submandibular gland cells. Some BMSCs in co-cultured cells showed submandibular gland acinar cell morphology. The proliferation rate of co-cultured cells was higher than that of submandibular gland cells (P <0.01), but no significant difference compared with normal submandibular gland cells and BMSCs (P> 0.05 ). Conclusion BMSCs have the potential to transform into submandibular gland acinar cells, and play a role in the repair of radioactive salivary gland cell injury.
