目的对接种麻疹减毒活疫苗(Measles Attenuated Live Vaccine,MV)后第9d出现的一例麻疹样病例标本,进行疫苗株和野毒株麻疹病毒鉴定。方法采用两种逆转录-聚合酶链反应-限制性片段长度多态性分析(Reverse Transcription-Polymerase Chain Reaction-Restriction Fragment Length Polymorphism,RT-PCR-RFLP)方法,检测麻疹样病例咽拭子标本。结果应用H1基因型麻疹野病毒RT-PCR-RFLP方法,对该病例咽拭子标本进行鉴定,其RT-PCR阳性产物经SalI酶切后,被切成两个片段,分别为408个和159个核苷酸,显示该病例是由H1基因型麻疹野病毒引起的。应用中国MV株RT-PCR-RFLP方法对该病例咽拭子标本进行鉴定,其RT-PCR产物不能被AflⅡ酶切,仍呈单一条带,显示该病例是由非MV株病毒引起的。结论接种MV第9d后出现的麻疹样病例,被证实为H1基因型麻疹野病毒引起,而非MV株病毒引起。
OBJECTIVE: To detect the measles-like cases in measles Attenuated Live Vaccine (MV) on the 9th day after vaccination with Measles Attenuated Live Vaccine (MV). Methods Two types of throat swab specimens were detected by RT-PCR-RFLP and reverse transcriptase-polymerase chain reaction-restriction fragment length polymorphism (RT-PCR-RFLP) Results The throat swab specimens were identified by RT-PCR-RFLP method. The RT-PCR positive products were digested with SalI and cut into two fragments, 408 and 159 Nucleotide, indicating that the case is caused by the H1 genotype measles wild virus. The throat swab specimens were identified by RT-PCR-RFLP in Chinese MV strains. The RT-PCR products could not be digested with Afl Ⅱ and remained single band, indicating that the cases were caused by non-MV strains. Conclusion The measles-like cases that appeared on the 9th day after inoculation of MV were confirmed to be caused by the wild type H1 measles virus rather than the MV strain.
