目的:观察蒙药肉蔻-5味丸对慢性抑郁模型大鼠行为及海马单胺递质的影响,探讨其抗抑郁作用机制。方法:60只雄性SD大鼠,随机分为正常组、模型组、盐酸氟西汀组(3.6 mg·kg-1)及肉蔻-5味丸低、中、高剂量组(0.2,0.4,0.8g·kg-1),每组10只。采用慢性不可预见性应激与孤养结合的造模方法复制抑郁模型,造模同时ig给药,每天1次,连续给药21 d。以体重变化,矿场试验(open-field test),糖水消耗试验来进行行为学评价,酶联免疫法(ELISA)检测大鼠海马5-羟色胺(5-HT),去甲肾上腺素(NE),多巴胺(DA)单胺递质的含量。结果:给予21 d应激后,模型组大鼠体重增长缓慢,糖水消耗量、Open-field试验水平活动得分和垂直活动得分及海马5-HT,DA,NE的含量较正常组显著降低(P<0.05,P<0.01),与模型组相比氟西汀和肉蔻-5味丸可不同程度改善其抑郁状态,氟西汀组、肉蔻-5味丸高、中剂量组大鼠海马5-HT,DA,NE的含量显著增加(P<0.05,P<0.01)。结论:肉蔻-5味丸的抗抑郁作用可能与提高脑内5-HT,NE,DA单胺递质的含量相关。
OBJECTIVE: To observe the effect of Mongolian medicine Kou Kou-5 Wei Wan on behavior and monoamine neurotransmitter in hippocampus of rats with chronic depression, and to explore its mechanism of antidepressant effect. Methods: Sixty male Sprague-Dawley rats were randomly divided into three groups: normal group, model group, fluoxetine hydrochloride group (3.6 mg · kg -1) and low, middle and high dose of nutmeg-flavored pills group (0.2, 0.4, 0.8g · kg-1), 10 in each group. The model of chronic unpredictable stress combined with solitary and nociception was used to replicate the model of depression. The model was given ig once a day for 21 days. The behavioral evaluation was carried out by weight change, open-field test and sugar consumption test. Serum levels of serotonin (5-HT) and norepinephrine (NE) in the hippocampus of rats were detected by enzyme-linked immunosorbent assay , Dopamine (DA) monoamine neurotransmitter content. Results: After 21 d stress, the weight gain of rats in the model group was slowed down. The levels of water consumption, open-field activity and vertical activity and the levels of 5-HT, DA and NE in the hippocampus were significantly lower than those in the normal group <0.05, P <0.01). Compared with the model group, fluoxetine and nutmeg-flavored pills could improve the depression of the rats in varying degrees, and hippocampus of fluoxetine, The content of 5-HT, DA and NE increased significantly (P <0.05, P <0.01). Conclusion: The antidepressant effect of the nutmeg-flavored pill may be related to increasing the content of 5-HT, NE, DA monoamine neurotransmitters in the brain.
