目的:研究红景天苷对缺氧条件下H9C2心肌细胞凋亡及Caspase-3表达的影响。方法:建立心肌细胞缺氧模型,本实验将细胞分为正常对照组、低氧组、低氧加红景天苷组(低剂量10mg/L、中剂量50mg/L、高剂量100mg/L),按实验设计因素处理后,用MTT检测取对数生长期细胞。TUNEL染色观察细胞凋亡,Westernblot测定Caspase-3表达。结果:与低氧组比较,红景天苷组各组均可明显提高缺氧损伤的H9C2心肌细胞存活率(P<0.01),降低细胞凋亡率(P<0.01),可有效减少缺氧H9C2心肌细胞内Caspase-3的表达(P<0.01)。结论:红景天苷可通过提高低氧条件H9C2心肌细胞存活率,降低细胞凋亡率及下调Caspase-3,从而保护心肌细胞。
Objective: To study the effect of salidroside on the apoptosis and the expression of Caspase-3 in H9C2 myocardial cells under hypoxia conditions. Methods: The model of cardiomyocyte hypoxia was established. The cells were divided into normal control group, hypoxia group, hypoxia plus salidroside group (low dose 10mg / L, medium dose 50mg / L, high dose 100mg / L) , According to the experimental design factors, using MTT detection of logarithmic growth phase cells. Cell apoptosis was observed by TUNEL staining and Caspase-3 expression was detected by Western blot. Results: Compared with the hypoxia group, all the groups of salidroside could obviously improve the hypoxia-injured H9C2 cardiomyocyte survival rate (P <0.01) and reduce the apoptosis rate (P <0.01), which could effectively reduce the hypoxia Caspase-3 expression in H9C2 cardiomyocytes (P <0.01). Conclusion: Salidroside can protect cardiomyocytes by increasing the survival rate of hypoxic H9C2 cardiomyocytes, decreasing the rate of apoptosis and decreasing Caspase-3.
