以水稻根尖和花药为材料,采用压片法和涂片法对水稻有丝分裂制片和减数分裂制片效果进行了比较研究。结果表明,根尖在常温下1N盐酸20min和45%醋酸处理与60℃水浴下1N盐酸10min+45%醋酸处理后压片效果较好,但压片法不及涂片法;花药在常温下1N盐酸15min+45%醋酸处理与60℃水浴下1N盐酸5~7.5min+45%醋酸处理后压片效果较好,且压片法适于减数分裂的染色体制片。
The effects of mitosis preparation and meiosis on rice root tip and anther were compared using tabletting method and smear method. The results showed that the rooting tips were better than 1N hydrochloric acid at normal temperature for 20min and 45% acetic acid treatment and 1N hydrochloric acid at 60 ℃ for 10min and 45% acetic acid treatment, but the tabletting method was inferior to smear method. % Acetic acid treatment and 60 ℃ water bath under the 1N hydrochloric acid 5 ~ 7.5min + 45% acetic acid after the tabletting effect is better, and the tabletting method for meiosis chromosome preparation.
