目的检测三阴乳腺癌组织(TNBC)中Notch1、AKT1和eIF4E表达并探讨其意义。方法采用免疫组织化学S-P法检测56例TNBC组织和30例癌旁正常乳腺组织Notch1、AKT1和eIF4E的表达情况。结果 TNBC组织中Notch1、AKT1和eIF4E阳性表达定位于细胞质和(或)细胞核,其表达率分别为62%(35/56)、57%(32/56)和61%(34/56),明显高于癌旁正常组织(P<0.01);56例TNBC组织中,Notch1、AKT1和eIF4E蛋白在组织学分级Ⅲ级阳性表达率高于Ⅰ+Ⅱ级(P<0.05),Notch1、AKT1和eIF4E的病理分期Ⅲ~Ⅳ期的表达率高于Ⅰ~Ⅱ期(P<0.05),Notch1、AKT1和eIF4E阳性表达与淋巴结转移有关(P<0.05);相关性分析显示,三者之间呈正相关(P<0.05)。Cox回归多因素分析显示Notch1和eIF4E的阳性表达是独立预后因素。结论 Notch1、AKT1和eIF4E在TNBC中呈明显高表达,与TNBC的发生、发展和转移密切相关。
Objective To detect the expression of Notch1, AKT1 and eIF4E in triple negative breast cancer (TNBC) and explore its significance. Methods Immunohistochemical S-P method was used to detect the expression of Notch1, AKT1 and eIF4E in 56 cases of TNBC and 30 normal breast tissues. Results The positive expression of Notch1, AKT1 and eIF4E in TNBC was located in the cytoplasm and/or nucleus. The expression rates of Notch1, AKT1 and eIF4E were 62% (35/56), 57% (32/56) and 61% (34/56) respectively. Higher than adjacent normal tissue (P<0.01); In 56 TNBC tissues, the positive rate of Notch1, AKT1 and eIF4E protein in histological grade III grade was higher than that of grade I+II (P<0.05), Notch1, AKT1 and eIF4E The positive expression rate of Notch1, AKT1 and eIF4E was significantly correlated with the lymph node metastasis (P<0.05). The correlation analysis showed that there was a positive correlation between the three. (P<0.05). Cox regression multivariate analysis showed that positive expression of Notch1 and eIF4E were independent prognostic factors. Conclusion Notch1, AKT1 and eIF4E are highly expressed in TNBC, which is closely related to the occurrence, development and metastasis of TNBC.
