目的 研究肝细胞癌细胞凋亡和浸润转移潜能的关系。方法 肝细胞癌 5 7例 ,用流式细胞仪检测细胞凋亡率 (AR)和细胞增殖活性 [用S期细胞比率 (SPF)和增殖指数 (PI)表示 ] ,用链霉亲合素 生物素酶复合物免疫组织化学检测 p2 1、p170和ndpk。 结果 AR (1.77± 0 .19) % ,SPF(12 .5 5± 0 .68) % ,PI(2 0 .91± 1.12 ) % (r =-0 .173 ) ;AR≤ 1.77%者 p170 (+ )、p2 1(+ )、ndpk(+ )分别为 61.3 6%、68.18%、5 2 .2 7% ,AR >1.77%者p170 (+ )、p2 1(+ )、ndpk (+ )分别为2 3 .0 8%、3 8.46%、84.62 % (P均 <0 .0 5 ) ;浸润转移组ndpk(+ ) 43 .75 % ,p2 1(+ ) 75 .0 0 % ,p170(+ ) 65 .63 % ,AR(1.12± 0 .16) % ,PI(2 3 .78± 1.48) % ,SPF(13 .90± 0 .99) % ,非浸润转移组ndpk(+ ) 80 .0 0 % ,p2 1(+ ) 44 .0 0 % ,p170 (+ ) 3 6.0 0 % ,AR (2 .3 2± 0 .5 2 ) % ,PI(18.5 3± 0 .82 ) % ,SPF(11.43± 0 .70 ) % (P均 <0 .0 5 )。结论 肝细胞癌细胞凋亡与其浸润转移潜能呈负相关 ,且与细胞增殖活性、ndpk、p2 1和 p170等多因素有关。
Objective To study the relationship between hepatocellular carcinoma cell apoptosis and invasion and metastasis potential. Methods Fifty-seven patients with hepatocellular carcinoma (HCC) were examined for apoptotic rate (AR) and cell proliferation by flow cytometry (S phase cell ratio (SPF) and proliferation index (PI) The enzyme complex was used to detect p2 1, p170 and ndpk by immunohistochemistry. Results AR (1.77 ± 0.19)%, SPF (12.5 ± 0.68)%, PI (20.91 ± 1.12)% (r = -0.137) P1 (+), p2 1 (+) and ndpk (+) were 61.36%, 68.18%, 52.27% and AR> 1.77% Were 23.0%, 23.46% and 84.62%, respectively (P <0.05); the infiltration and metastasis groups were 43.75% ) In the non-invasive metastasis group was significantly higher than that in the non-invasive group (65.63%, AR 1.12 ± 0.16%, PI 23.78 ± 1.48%, SPF 13.90 ± 0.99% 0%, p2 1 (+) 44.0%, p170 (+) 3 6.0 0%, AR (2.3.2 ± 0.52)%, PI 11.43 ± 0.70)% (all P <0.05). Conclusion The apoptosis of hepatocellular carcinoma cells is negatively correlated with the invasiveness and metastasis potential of hepatocellular carcinoma cells, and is related to many factors such as cell proliferation activity, ndpk, p21 and p170.
