目的:研究地黄饮子水煎剂对去势大鼠骨强度和骨病理形态结构的影响,进一步探讨其作用机制。方法:采用去卵巢法制备大鼠骨质疏松症模型,将大鼠随机分为假手术组,模型组,地黄饮子水煎剂低、中、高剂量组,西药组(戊酸雌二醇片),每组10只,分别予相应药物灌胃4周,股动脉采血处死大鼠。观察大鼠的一般状况,称量体重,比色分析法检测血清钙(calcium,Ca),磷(phosphorus,P)含量,酶联免疫吸附法(ELISA)检测血清骨钙素(osteocalcin,BGP)含量和抗酒石酸酸性磷酸酶(tartrate resistant acid phosphatase,tr ACP)含量,双能X射线吸收测量仪平台上测定右股骨部位骨密度(bone mineral density,BMD),用AG-IX生物力学万能实验机检测股骨最大载荷,定位-伸长位移及苏木素-伊红(HE)染色法观察股骨的病理形态学变化。结果:与假手术组比较,模型组大鼠BMD,Ca,P,BGP,最大载荷,定伸长-位移均显著降低(P<0.05),而模型组大鼠体重和tr ACP的含量及地黄饮子水煎剂各干预组大鼠体重则显著增加(P<0.05),同时模型组骨小梁结构不完整、丢失、断裂严重。与模型组相比,地黄饮子水煎剂各干预组大鼠BMD,BGP,Ca,最大载荷,定伸长-位移均显著升高(P<0.05),其高、中剂量组大鼠P含量显著升高,而其高剂量组大鼠体重和tr ACP的含量则显著降低(P<0.05),并且地黄饮子水煎剂高剂量组骨小梁排列规则,数目增多、增宽。结论:地黄饮子能够提高绝经后骨质疏松大鼠的骨密度,促进骨形成、抑制骨吸收,对绝经后骨代谢异常具有积极的防治作用。
Objective: To study the effect of Decoction of Decoction of Rehmanniae Decoction on bone strength and pathomorphology of ovariectomized rats, and to explore its mechanism. Methods: Osteoporosis model was established by ovariectomized rats. Results: Compared with the sham operation group, the maximal load, fixed elongation and displacement of BMD, Ca, P, BGP in model group were significantly decreased (P <0.05), while the body weight and tr ACP content in model group were The body weight of rats in each intervention group was significantly increased (P <0.05), meanwhile, trabecular structure was incomplete, lost and severed in the model group. Compared with model group, the maximal load, fixed elongation and displacement of BMD, BGP and Ca in rats of each group were significantly increased (P <0.05), and the contents of P (P <0.05), and the high dose group of high dose group Rehmanniae Decoction of high dose trabecular arrangement of rules, the number increased, broadening. Conclusion: Rehmanniae Decoction can improve bone mineral density, promote bone formation and inhibit bone resorption in postmenopausal osteoporosis rats, and have a positive effect on preventing and treating postmenopausal bone metabolism.
