采用木瓜蛋白酶(papain)和碱性蛋白酶(Alcalase 2.4L)分别水解新疆产背暗异唇蚓(Allolobophora caliginosa trapezoides, Duges 1828)的蛋白,以氨基酸含量为衡量指标,通过单因素与正交试验设计,对最佳酶条件进行比较筛选研究,并分析了酶解液氨基酸含量。结果表明:采用木瓜蛋白酶最佳酶解条件为:温度 50℃、pH7.5、酶浓度 0.3%、作用时间 8h,而碱性蛋白酶的最佳酶解条件为:温度 50℃、pH7、酶浓度 0.3%、作用时间 8h,在此条件下,酶解背暗异唇蚓蛋白的水解液氨基酸含量分别可达到16.55mg/ml 和29.48mg/ml,Alcalase 2.4L蛋白酶对背暗异唇蚓的酶解效率比木瓜蛋白酶高。因此,需要制造新型富含氨基酸、微量元素及小肽添加剂时,可选用Alcalase 2.4L。
Proteins from Allolobophora caliginosa trapezoides (Duges 1828) were hydrolyzed by papain and Alcalase 2.4L, respectively. The amino acid content was used as a measure. Alcalase 2.4L Solution efficiency higher than papain. Therefore, the need to create a new rich amino acids, trace elements and small peptide additives, the choice of Alcalase 2.4L.
