本文用细胞培养的方法评价了外周血干细胞移植(PBSCT)后一年造血系统损伤的情况,并明确这种损伤是发生于造血祖细胞水平还是基质细胞水平。 方法 PBSCT后一年的患者共31例,其中乳腺癌患者9例,非霍奇金淋巴瘤17例,霍奇金病(HD)5例。用三种细胞培养法,即CFU-GM集落形成分析,塑料粘附祖细胞△分析法(P△)和长期骨髓培养(LTBMC)。用Ficoll-Hypaque梯度离心法分离单个核细胞供CFU-GM集落形成试验用和p△试验用,用引力沉降分离骨髓细胞供LTBMC用。正常对照共48例,包括
In this paper, the cell culture method was used to evaluate the hematopoietic system damage after one year of peripheral blood stem cell transplantation (PBSCT), and it was determined whether this injury occurred at the level of hematopoietic progenitor cells or stromal cells. Methods A total of 31 patients were included in one year after PBSCT, including 9 cases of breast cancer, 17 cases of non-Hodgkin’s lymphoma, and 5 cases of Hodgkin’s disease (HD). Three cell culture methods were used, namely CFU-GM colony formation assay, plastic adhesion progenitor cell delta assay (P[Delta]) and long-term bone marrow culture (LTBMC). Mononuclear cells were isolated by Ficoll-Hypaque gradient centrifugation for CFU-GM colony formation assay and pΔ assay, and bone marrow cells were isolated by gravity sedimentation for LTBMC. A total of 48 normal controls were included.
