采用细胞培养技术及MTT法,观察培养状态下脊索瘤细胞的生长特点及化疗药物对脊索瘤细胞生长的抑制作用.方法:手术切除的新鲜脊索瘤标本,置入含IOOmL/L小牛血清的RPMI164O培养基中,观察脊索瘤细胞生长特点;取对数生长期的细胞接种96孔板,每孔细胞数1o‘个.将质量浓度3,3O,75mg/L顺铂;15,15o,375mg门。卡钥;l,Ic,25ms/l。表阿霉素分别加入孔内,37CSOmL/LCO。培养箱中培养48h,用MTT法计算抑制率.结果:培养状态下的脊索瘤细胞大小不一,形态不规则,很多的细胞质内有泡状突起,核分裂像较多见,贴壁生长,但贴壁发生较慢.顺铂、卡铂对脊索瘤细胞生长的抑制率相同,在9%~22%之间,随质量浓度增加其抑制率增加.而表阿霉素对脊索瘤细胞生长几无抑制作用.结论:常用的化疗药物顺钥、卡铂对脊索瘤细胞生长有一定的抑制作用.而表阿霉素对其生长无抑制作用.
Cell culture technique and MTT method were used to observe the growth characteristics of chordoma cells in culture state and the inhibitory effect of chemotherapeutic drugs on the growth of chordoma cells. METHODS: Fresh chordoma specimens surgically resected were placed in RPMI164O medium containing 100 mL/L calf serum to observe the growth characteristics of chordoma cells. Cells in logarithmic growth phase were seeded on 96-well plates, and the number of cells per well was 1o’. One. The mass concentration of 3,3O, 75mg/L cisplatin; 15,15o, 375mg gate. Card key; l, Ic, 25ms/l. Epirubicin was added to the wells, 37 CSOmL/LCO. The incubator was cultured for 48 hours and the inhibition rate was calculated by MTT method. RESULTS: The chordoma cells in culture were of different size and irregular morphology. Many cytoplasms had vesicular processes. The mitoses were seen more often and adhered to the wall, but the adherence was slower. Cisplatin and carboplatin had the same inhibitory effect on the growth of chordoma cells, ranging from 9% to 22%. The inhibition rate increased with increasing mass concentration. The epirubicin did not inhibit the growth of chordoma cells. Conclusion: The commonly used chemotherapeutic drugs, cis-key and carboplatin, have a certain inhibitory effect on the growth of chordoma cells. Epirubicin has no inhibitory effect on its growth.
