目的探讨不同浓度粉防己碱对离体豚鼠心脏缺血时去甲肾上腺素(NE)释放及其在轴浆内代谢的影响。方法用高效液相色谱-电化学法测定豚鼠离体心脏灌注流出液中NE和二羟基苯基乙二醇(DHPG)的含量,将正常灌注下(S1)和不同条件缺血灌注下(S2)两刺激进行自身前后对比。结果各浓度组S2刺激期间NE的释放量较缺血对照组明显减少(P<0.05),且各浓度组间均有显著性差异(P<0.05);DHPG的量在缺血对照组和低浓度组间并无显著性差异(P>0.05),而中浓度和高浓度组DHPG的量较对照组和低浓度组有了明显升高(P<0.05),且高浓度组与中浓度组间有显著性差异(P<0.05)。结论粉防己碱可以明显抑制缺血时离体豚鼠心脏NE的释放,其作用随浓度升高而增强,而较高浓度的粉防己碱在抑制NE释放的同时还增强了轴浆中NE的代谢,且此作用也随浓度升高而增强。
Objective To investigate the effect of different concentrations of tetrandrine on the release of norepinephrine (NE) and its effect on axonole metabolism in isolated guinea pig hearts. Methods The contents of NE and dihydroxyphenyl glycol (DHPG) in the pericardial perfusion effluent of guinea pigs were determined by high performance liquid chromatography-electrochemical detection. There was a significant difference between them (P<0.05). Conclusion Tetrandrine can significantly inhibit the release of NE from isolated hearts of guinea pigs during ischemia. The effect of tetrandrine increases with the increase of concentration. The higher concentration of tetrandrine can inhibit the release of NE and increase the metabolism of NE in axons. , and this effect also increases with increasing concentrations.
