以不同来源北冬虫夏草为试材,通过正交实验研究总黄酮的提取工艺,以紫外吸收峰的峰形、位置及强度为标准选定显色试剂及测定波长,确定北冬虫夏草中总黄酮的提取工艺及含量测定方法。结果表明:用40倍量的70%乙醇于90℃水浴中单次回流提取4h,总黄酮提取率最高;以芦丁为对照品,0.1mol/L的AlCl3为显色剂,265nm为检测波长,测定总黄酮的含量方法最佳;以大米为培养基的北冬虫夏草(米虫草),蚕蛹培养(蛹虫草)的北冬虫夏草子实体、全草及虫体中总黄酮含量分别为3.700 8、4.039 8、3.219 2、2.502 4mg/g。
