【目的】研究一株黄蓝状菌(Talaromyces flavus)解磷能力和解磷条件的优化,为解磷菌肥的开发提供优良菌株和培养技术。【方法】实验选取一株从杨树根部分离的促生真菌菌株SH16,分别采用钼锑抗比色法和钒钼酸铵法测定其解磷能力,以磷酸钙为磷源,测定不同培养因素对解磷效果的影响。【结果】菌株SH16对磷酸钙、过磷酸钙、磷酸铝、植酸钙等4种磷源均具有一定的溶解作用,对磷酸钙溶解效果最好,7 d后可溶性磷浓度达660.9 mg/L,解磷效果可达45 U/L;初始p H、碳源、温度及Na Cl质量分数对菌株SH16的解磷效果影响最大。设定初始p H为8,以葡萄糖为碳源,培养温度为30℃,培养基中添加50 mg/L的水溶性磷时,该菌株的解磷效果最好。菌株对Na Cl具有较强的耐受性,随着Na Cl质量分数升高,解磷量逐渐下降,当Na Cl质量分数达到5%时,解磷量仍可达173.1 mg/L。【结论】菌株SH16对磷酸钙表现出较高的溶解活性,可以进一步开发为生物菌肥。
【Objective】 The objective of this study was to optimize the conditions for the solution of one strain of Talaromyces flavus and the conditions for the solution of phosphorus, and to provide excellent strains and culture techniques for the development of phosphate-solubilizing bacterial fertilizer. 【Method】 A strain of protozoa fungal strain SH16 isolated from the roots of poplar was selected in this experiment. The antiphospholysis ability was determined by molybdenum-antimony colorimetric method and ammonium vanadate molybdate method respectively. Calcium phosphate Effect on Phosphorus Removal. 【Result】 Strain SH16 had certain dissolution effects on four kinds of phosphate sources such as calcium phosphate, superphosphate, aluminum phosphate and calcium phytate, and had the best effect on calcium phosphate dissolution. After 7 days, the soluble phosphorus concentration reached 660.9 mg / L , Phosphorus up to 45 U / L; initial p H, carbon source, temperature and Na Cl mass fraction had the greatest effect on the phosphate solubilization of strain SH16. The initial pH was set to 8, glucose as a carbon source, the culture temperature was 30 ℃, the medium when adding 50 mg / L of water-soluble phosphorus, the strain of the best phosphorus solution. The strain was more tolerant to NaCl. With the increase of NaCl concentration, the amount of phosphate solubilizing gradually decreased. When NaCl mass fraction reached 5%, the amount of phosphate solubilizing still reached 173.1 mg / L. 【Conclusion】 Strain SH16 showed high activity for calcium phosphate and could be further developed into bio-fertilizer.
