运用化学渗透法提取藻体(浒苔)中的谷胱甘肽(GSH),利用邻苯二甲醛和GSH反应生成的荧光体系,在激发波长340 nm,发射波长420 nm条件下,用荧光分光光度计进行测定,建立了快速测定藻体中GSH含量的方法。结果表明,GSH质量浓度在0.00~1.60 mg/L范围内线性较好,相关性系数r=0.9980,检出限为0.03 mg/L,回收率在98.2%~101.0%之间,样品与标准品测定的相对标准偏差分别为0.63%和0.26%;实验中利用不同浓度Cd2+诱导海带,考察了海带对Cd2+的富集能力及GSH变化趋势,并研究了Cd浓度与GSH变化相关性,结果证明海带对Cd富集浓度较高,其Cd含量变化与GSH变化趋势呈现较好的正相关性;同时,利用该方法测定了青岛6个不同区域的浒苔中GSH含量,结果发现不同区域浒台中GSH含量有轻微差异。
The glutathione (GSH) in algae (Enteromorpha prolifera) was extracted by chemical permeation. Fluorescence spectroscopy was performed using the fluorescence system formed by the reaction of o-phthalaldehyde and GSH at the excitation wavelength of 340 nm and the emission wavelength of 420 nm. Photometer was measured to establish a rapid determination of GSH content in algae. The results showed that the linearity of GSH concentration ranged from 0.00 to 1.60 mg / L with a correlation coefficient of r = 0.9980. The detection limit was 0.03 mg / L and the recoveries ranged from 98.2% to 101.0% The relative standard deviations (RSDs) were 0.63% and 0.26%, respectively. Cd2 + -induced kelp was used to study the enrichment ability of Cd2 + and the trend of GSH, and the correlation between Cd concentration and GSH was also studied. The results showed that there was a positive correlation between the change of Cd content and the change trend of GSH in Cd concentration. Meanwhile, the GSH content of Enteromorpha in six different regions of Qingdao was determined by this method. It was found that GSH There is a slight difference in content.
