目的 研究分化诱导剂六亚甲基二乙酰胺 (hexamethylenebisacetamide,HMBA)对人粘液表皮样癌细胞系MEC 1体外粘附、移动和侵袭的影响。方法 用 2mmol/LHMBA体外诱导培养MEC 1细胞 ,用MTT(thiazalylblue ,噻唑蓝 )比色法测定细胞在重构基底膜Matrigel上的粘附 ,用Millicell PCF培养系统测定细胞的移动性和侵袭性。结果 HMBA诱导的MEC 1细胞在 30min和 90min粘附实验中的粘附率 ,分别下降了 2 6 81%和 31 13% (P <0 0 5 )。HMBA对MEC 1细胞体外移动和侵袭的抑制率分别为 6 3 49%和 6 7 6 5 % (P <0 0 5 )。结论 HMBA对人粘液表皮样癌细胞MEC 1具有抗侵袭作用
Objective To study the effect of differentiation-inducing agent hexamethylenebisacetamide (HMBA) on the adhesion, migration and invasion of human mucoepidermoid carcinoma cell line MEC 1 in vitro. Methods MEC 1 cells were induced by 2 mmol / LHMBA in vitro. The adhesion of the cells to the reconstituted basement membrane Matrigel was measured by thiazalyl blue (MTT) colorimetric assay. The cell migration and invasion were measured by Millicell PCF culture system. Results The adhesion rate of HMBA-induced MEC 1 cells in the adhesion experiments of 30 min and 90 min decreased by 2 6 81% and 31 13%, respectively (P 0 05). The inhibitory rates of HMBA on MEC 1 cells in vitro migration and invasion were 6 3 49% and 6 7 6 5%, respectively (P 0 05). Conclusion HMBA has an anti-invasive effect on human mucoepidermoid carcinoma cell line MEC 1
