在NADPH等辅助因子的参与下,一氧化氛合酶(NOS)能催化L-精氨酸生成一氧化氮(NO)。NO与过氧化氢反应生成过氧亚硝酸,过氧亚硝酸能与鲁米诺产生化学发光反应,当NO浓度为100fmol/L~Inmol/L时,发光强度与NO量成正比.根据这一原理、利用化学发光分析建立了一种新的NOS活性测定方法。用该方法测量10只大鼠脑组织中的NOS活性,其NOS活性为28.3±6.9pmolNO/mg蛋白/min。
In the presence of cofactors such as NADPH, nitric oxide synthase (NOS) catalyzes the production of nitric oxide (NO) by L-arginine. Nitric oxide reacts with hydrogen peroxide to produce peroxynitrite. Peroxynitrite can react with luminol to produce chemiluminescence reaction. When the concentration of NO is 100 fmol / L ~ Inmol / L, the luminescence intensity is proportional to the amount of NO. Based on this principle, a new method for determination of NOS activity was established by chemiluminescence analysis. NOS activity in brain tissue of 10 rats was measured by this method and its NOS activity was 28.3 ± 6.9 pmolNO / mg protein / min.
