应用不同浓度的C-myc反义寡核苷酸处理体外培养的经10-8mol/L内皮素-1(ET-1)诱导的人肺动脉平滑肌细胞,通过细胞计数和3H-TdR掺入值观察该反义核酸对细胞增殖及DNA合成的抑制作用,应用斑点杂交法检测平滑肌细胞C-fos和C-mycmRNA的表达水平。结果表明,10、20、50μg/ml反义寡核苷酸对细胞计数和3H-TdR掺入值与对照组相比抑制率分别为11.9%和12.3%(P<0.05),21.6%和14.7%(P<0.01),36.3%和27.2%(P<0.01);对C-mycmRNA表达抑制率分别为71.3%(P<0.05),80.7%(P<0.05),92.5%(P<0.01);50μg/ml反义核酸对C-fosmRNA表达抑制率是79.7%,10、20μg/ml组无抑制作用。C-myc反义核酸能显著抑制内皮素诱导的人肺动脉平滑肌细胞增殖,其作用系依赖反义核酸的序列特异性和剂量依赖。
