观察高糖对体外培养肾小球系膜细胞表达纤维连接蛋自 (FN)、纤溶酶原激活物抑制物(PAI 1)mRNA以及细胞增殖、分泌FN的影响 ,同时观察益肾化浊注射液对高糖引起上述各指标变化的影响。采用体外培养肾小球系膜细胞方法 ,用高糖及益肾化浊注射液作为刺激物 ,四甲基偶氮唑盐微量酶比色法 (MTT)检测细胞增殖 ,酶联免疫吸附测定法 (ELISA)测定细胞上清FN含量 ,Northem杂交检测FN、PAI 1mRNA表达水平。结果表明 ,高糖可促进分泌FN ,上调PAI 1mRNA的表达 ,明显抑制系膜细胞的增殖 ;而益肾化浊注射液可缓解高糖所引起的上述指标的改变 ,并可下调FNmRNA的表达。
To observe the effect of high glucose on the expression of fibronectin (FN), plasminogen activator inhibitor (PAI 1) mRNA, cell proliferation, and FN secretion in cultured glomerular mesangial cells in vitro, and observe the Yishen Huazhuo injection The effect of liquid on high glucose causes changes in the above indicators. In vitro culture of glomerular mesangial cells was performed using high glucose and Yishen Huazhuo injection as stimulators. MTT assay was used to detect cell proliferation. ELISA was used to determine the FN content in the supernatant of the cells, and Northern hybridization was used to detect the expression of FN and PAI 1 mRNA. The results showed that high glucose can promote the secretion of FN, up-regulate the expression of PAI 1 mRNA, and significantly inhibit the proliferation of mesangial cells; and Yishen Huazhuo injection can alleviate the above-mentioned changes caused by high glucose, and can down-regulate the expression of FN mRNA.
