目的:采用RAPD技术,对积雪草DNA分子标记进行研究。方法:以不同来源的积雪草为样本提取基因组DNA,通过优化PCR反应体系,选择适合的随机引物,扩增分析积雪草的特征条带。结果:经优化反应体系扩增,获得不同来源及不同生长环境的积雪草特异的种群条带,具有高度稳定的遗传特征。积雪草的种群特征带与绞股蓝、烟草、乌蔹莓等其他不同科属的植物有明显的区别。结论:所建立的RAPD分析方法扩增不同来源的积雪草显示高度的均一性,可用于积雪草真品和混淆品的鉴别。
Objective: To study the molecular markers of Centella asiatica using RAPD technology. Methods: The genomic DNA was extracted from Centella asiatica from different sources. By optimizing the PCR reaction system and selecting suitable random primers, the characteristic bands of Centella asiatica were amplified and analyzed. RESULTS: After optimization of the reaction system, A. asiatica-specific population bands from different sources and different growth environments were obtained, with highly stable genetic characteristics. The population characteristics of Centella asiatica are obviously different from those of other plants of different families, such as Gynostemma pentaphyllum, tobacco and black berry. Conclusion: The established RAPD analysis method can amplify the degree of homogeneity of Centella asiatica from different sources and can be used to identify the authenticity and confusion of Centella asiatica.
