为分析化学诱变产生的玉米细胞质雄性不育材料的不育类型,采用田间育性调查、花粉粒活力检测、胞质不育特异引物PCR扩增、小斑病菌抗性鉴定以及石蜡切片等方法来分析不育系85218A的胞质不育类型。结果表明,不育系85218A田间败育彻底,I2-KI染色后花粉粒呈淡黄色,属于典型的圆败,PCR扩增为C型不育系,对C型小斑病菌表现中抗,花粉粒败育关键时期为单核后期。该研究拓宽了我国玉米雄性不育种质资源。
In order to analyze the types of sterility of maize cytoplasmic male sterile (CMS) material generated by chemical mutagenesis, field fertility investigation, pollen viability test, PCR amplification of cytoplasmic male sterility-specific primers, resistance to P. xylostella and paraffin section To analyze the CMS type 85218A. The results showed that the male sterile line 85218A was aborted completely, and the pollen grains were pale yellow after I2-KI staining. It belongs to the typical round failure, and was PCR-amplified to C-type sterile line. The critical stage of abortion is mononuclear stage. This study broadens the maize male sterile germplasm resources in China.
