目的 :研究普罗布考对受损内皮细胞增殖和再生内皮细胞功能修复的影响。方法 :分别采用MTT比色法和PCNA免疫组化法测定体外培养的人脐静脉内皮细胞所处增殖状态 ;以兔胸主动脉血管环对乙酰胆碱舒张反应和内皮细胞分泌NO、PGI2 的能力评价内皮细胞功能状况。结果 :LPC作用 2 4h可导致内皮细胞增殖显著受抑及分泌NO、PGI2 功能受损 ,不同剂量普罗布考 (2 0 ,4 0 ,80 μmol·L- 1)可促进LPC损伤的内皮细胞增殖 (5 5 .5 % ,5 9.3% ,72 .2 %vs 31.7% ,P <0 .0 1) ,同时提高血管成型术后兔胸主动脉血管环内皮依赖性舒张功能 (6 6 .6 3%vs 2 5 .95 % ,P <0 .0 1) ,并修复LPC损伤的内皮细胞分泌NO、PGI2 的功能 (P <0 .0 1)。结论 :普罗布考可促进受损内皮细胞增殖和再生内皮细胞功能修复。
OBJECTIVE: To study the effect of probucol on the proliferation of endothelial cells and the function of regenerated endothelial cells. Methods: The proliferative state of human umbilical vein endothelial cells cultured in vitro was determined by MTT colorimetry and PCNA immunohistochemistry respectively. The ability of acetylcholine relaxation and endothelial cell secretion of NO and PGI2 in rabbit thoracic aortic rings were evaluated by endothelium Cell function status. RESULTS: LPC treatment for 24 h resulted in marked inhibition of endothelial cell proliferation and impaired NO secretion, and PGI2 function was impaired. Different doses of probucol (20, 40, 80 μmol·L-1) promoted endothelial cell proliferation induced by LPC (55.5% vs59.3%, 72.2% vs31.7%, P <0.01), and at the same time increased the endothelium-dependent vasodilation of rabbit thoracic aortic vessels after angioplasty (66.36% % vs 25.95%, P <0.01), and restored the function of NO and PGI2 secreted by LPC-injured endothelial cells (P <0.01). Conclusion: Probucol can promote the proliferation of damaged endothelial cells and the function of regenerative endothelial cells.
