目的:探讨正丁酸钠(SB)对人肝癌细胞的生物学效应.方法:以1mmol/L的SB作用于培养的人肝癌细胞,观察肝癌细胞某些生物学行为的变化.结果:在SB作用下,人肝癌细胞生长抑制.细胞形态改变,ConA凝集现象消失,琼脂集落形成率下降(p<0.001),染色体主流范围变窄,二倍体及亚二倍体比例升高,~3H—TdR掺人率、细胞DNA含量下降与对照比较有显著差异(p<0.01).结论:人肝癌细胞在丁酸钠作用下,某些恶性生物学行为可发生部分逆转.
Objective: To investigate the biological effects of sodium butyrate (SB) on human hepatocellular carcinoma cells. METHODS: Human hepatocellular carcinoma cells cultured with 1 mmol/L SB were used to observe the changes of some biological behaviors of hepatoma cells. Results: In SB Under the action, human hepatocellular carcinoma cell growth was inhibited, cell morphology was changed, ConA agglutination disappeared, agar colony formation rate decreased (p<0.001), chromosome mainstream range narrowed, diploid and hypodiploid ratio increased, ~3H- The decrease of TdR incorporation rate and cell DNA content was significantly different from that of the control (p<0.01). Conclusion: Under the action of sodium butyrate, some malignant biological behaviors of human hepatoma cells may be partially reversed.
