目的观察血肿周边脑组织中HIF-lα和VEGF的表达情况并探讨二者表达的相关性。方法应用免疫组织化学技术、RT-PCR和Western蛋白印迹技术检测39例脑出血灶周脑组织中HIF-lα和VEGF的表达。结果脑出血灶周脑组织中HIF-lα、VEGF蛋白和mRNA的表达均随时间进行性增加,与正常脑组织组比较差异有统计学意义(P﹤0.01)。脑出血后不同时间HIF-lα、VEGF蛋白表达阳性率和mRNA分别比较,差异均有统计学意义(P﹤0.05);脑出血后HIF-1α和VEGF蛋白及mRNA的表达均呈正相关。结论 HIF-lα与VEGF在血肿周边脑组织中均有表达并具有一致性,可能与脑出血后微循环的重建有关。
Objective To observe the expression of HIF-1α and VEGF in the perihematomal brain tissue and to explore their correlation. Methods Immunohistochemistry, RT-PCR and Western blotting were used to detect the expression of HIF-1α and VEGF in 39 cases of cerebral hemorrhage. Results The expression of HIF-1α, VEGF protein and mRNA in the perihematoma brain tissue increased progressively with time, showing significant difference compared with normal brain tissue (P <0.01). The positive rates of HIF-1αand VEGF protein at different time after ICH were statistically different (P <0.05). There was a positive correlation between the expression of HIF-1αand VEGF protein and mRNA after intracerebral hemorrhage. Conclusions Both HIF-1α and VEGF are expressed in the brain tissue around the hematoma and are consistent, which may be related to the re-establishment of microcirculation after intracerebral hemorrhage.
