目的:研究红活麻总香豆素的制备工艺。方法:通过正交设计试验选择红活麻总香豆素的最佳提取条件,采用大孔吸附树脂对红活麻总香豆素进行纯化,紫外分光光度法测定总香豆素的含量。结果:红活麻总香豆素的最佳提取工艺为以4倍体积70%乙醇,提取3次,每次1.5 h。HPD300大孔树脂分离纯化红活麻总香豆素效果最佳,其工艺条件为:上样浓度0.4 g生药/mL,上样体积为30 mL,流速为2 BV/h,先用4 BV的蒸馏水除杂,再用4BV的70%乙醇洗脱;得率可达52%以上。结论:该方法适合于制备红活麻总香豆素,工艺的稳定性和重复性较好。
Objective: To study the preparation process of total coumarin in Honghumang. Methods: The optimal extraction conditions of total coumarin in Honghumang were selected by orthogonal design. The total coumarin in Honghumang was purified by macroporous adsorption resin and the total coumarin was determined by UV spectrophotometry. Results: The optimal extraction process of total coumarin was as follows: 4 times the volume of 70% ethanol, extracted three times, each 1.5 h. HPD300 macroporous resin separation and purification of red live cotatlonum best effect, the process conditions: the sample concentration of 0.4 g crude drug / mL, the sample volume of 30 mL, the flow rate of 2 BV / h, first with 4 BV Distilled in distilled water, and then 4BV 70% ethanol elution; Yield up to 52%. Conclusion: The method is suitable for the preparation of total red coumarin, the process of stability and repeatability is better.
