应用 DNA-RNA 斑点杂交方法研究了低碘鼠模型第15~18日龄胎鼠脑神经细胞原代培养物甲状腺激素受体(THR)c-erbAct mRNA 的表达。结果显示大脑神经细胞在0天时已可检测到 THRmRNA 的表达,培养第5天时表达水平略有增高,培养第10天时表达量大约可达80pg 左右,在培养第10~20天之间 THR mRNA 表达水平没有明显变化。在培养期间小脑神经细胞 THR mRNA 的表达水平较低,并且没有明显的变化。加入甲状腺激素(T_310~(-8)M)对脑神经细胞 THR 基因的表达没有明显的调节作用。与正常鼠相比低碘鼠脑神经细胞在成熟过程中 THR mRNA 的表达低于正常水平。推测这是IDD 脑损伤的一个重要环节。
The expression of c-erbAct mRNA of thyroid hormone receptor (THR) in primary cultured fetal rat neurons from the 15th to the 18th day in a low iodine mouse model was studied by DNA-RNA dot blot hybridization. The results showed that the expression of THR mRNA was detected in brain neurons at day 0, and the expression level was slightly increased on the fifth day after culture. The expression level of THR mRNA was about 80 pg at the 10th day after culture. THR mRNA expression No significant change in level. THR mRNA expression in cerebellar neurons was lower during culture, and there was no obvious change. The addition of thyroid hormone (T_310 ~ (-8) M) had no significant effect on THR gene expression in brain cells. Compared with normal mice, the expression of THR mRNA was lower than normal in brain cells of low iodine-deficient mice during maturation. Speculated that this is an important part of IDD brain injury.
