为估算经粒细胞集落刺激因子(G-CSF)动员的外周血中祖细胞和多能造血干细胞的含量,将不同数量的经G-CSF动员的外周血单个核细胞(PBMC)移植到经全身照射(TBI)的同系小鼠,与骨髓移植(BMT)组比较受体鼠外周血白细胞恢复动态及长期存活率。结果显示,输入相同剂量(1×106)细胞14和21天时,经动员的PBMC移植鼠的白细胞数远较BMT鼠为高(P<0.05),说明前者含有更多的祖细胞。150天存活期的观察发现,移植≥1×106经动员的PBMC或≥1×105骨髓细胞均可使80%左右的受体鼠长期存活;而移植1×105经动员的PBMC或1×104骨髓细胞均使50%受体鼠长期存活,提示经动员的外周血中多能造血干细胞的含量约为同样数目骨髓细胞的1/10。
To estimate the levels of peripheral blood progenitor cells and pluripotent hematopoietic stem cells mobilized by granulocyte colony-stimulating factor (G-CSF), different numbers of G-CSF-mobilized peripheral blood mononuclear cells (PBMCs) were transplanted into whole body (TBI), the recovery of peripheral blood leukocytes and the long-term survival of recipient mice were compared with that of bone marrow transplantation (BMT). The results showed that when the same dose (1 × 106) of cells was injected for 14 and 21 days, the number of leukocytes in mobilized PBMCs was much higher than that in BMT mice (P <0.05), indicating that the former contained more progenitor cells. 150-day survival of the observation found that the transplant ≥ 1 × 106 mobilized PBMC or ≥ 1 × 105 bone marrow cells can make about 80% of the recipient rat long-term survival; and transplanted 1 × 105 mobilized PBMC or 1 × 104 Bone marrow cells are 50% of the recipient mice long-term survival, suggesting that mobilized peripheral blood pluripotent hematopoietic stem cell content is about the same number of bone marrow cells 1/10.
