目的考察TGF-β1对人粘连成纤维细胞(ATF)增殖与COL-I合成的影响。方法通过人体腹膜粘连组织原代培养获取ATF,将不同剂量的TGF-β1与ATF共培养48 h,MTT法测定细胞增殖活性,ELISA法测定培养基中COL-I的含量。结果 TGF-β1 0.5~40 ng/mL剂量组的ATF增殖活性均高于0 ng/mL剂量组,差异有统计学意义(P<0.01)。TGF-β1 10、20 ng/mL剂量组的COL-I含量与0 ng/mL剂量组差异有统计学意义(P<0.01)。结论 TGF-β1可明显提高人ATF的增殖活性与COL-I合成水平。
Objective To investigate the effect of TGF-β1 on the proliferation of human adherent fibroblasts (ATF) and the synthesis of COL-I. Methods ATF was obtained through primary culture of human peritoneal adhesion tissue. Co-culture of different doses of TGF-β1 with ATF was performed for 48 hours. MTT assay was used to determine the cell proliferation activity. The content of COL-I in culture medium was determined by ELISA. Results TGF-β1 0.5 ~ 40 ng / mL dose group ATF proliferation activity were higher than the 0 ng / mL dose group, the difference was statistically significant (P <0.01). The concentration of COL-I in 10 ng / mL TGF-β1 group and 0 ng / mL dose group were significantly different (P <0.01). Conclusion TGF-β1 can significantly enhance the human ATF proliferation activity and COL-I synthesis level.
