目的:探讨IL6在系膜增殖性肾炎(MsPGN)发病中的作用。方法:采用逆转录聚合酶链式反应方法检测6例MsPGN、5例非MsPGN患儿肾活检组织及4例正常肾组织中IL6mRNA的相对表达水平。结果:MsPGN、非MsPGN患儿肾组织中IL6mRNA表达水平(x±s)分别为:0433±0111和0271±0023。MsPGN组IL6表达水平明显高于非MsPGN组(P<002)。4例正常肾组织中3例无IL6mRNA表达,余1例表达水平为0192。中度MsPGN和轻度MsPGN表达水平分别为:0491±0084和0317±0018,二者比值为15∶1,MsPGN伴肾小球局灶节段硬化和不伴硬化组其表达水平分别为0516±0083和0350±0058,二者比值为147∶1。另采用生物活性法测定MsPGN患儿尿中IL6活性水平,发现尿中IL6活性与肾组织中IL6mRNA的表达水平明显正相关(r=0977,P<0001)。结论:IL6可能与MsPGN发病机理有关,IL6可能参与MsPGN系膜增殖和硬化,尿中IL6活性测定可反映肾组织局部IL6mRNA的表达情况
Objective: To investigate the role of IL6 in the pathogenesis of mesangial proliferative glomerulonephritis (MsPGN). Methods: The relative expression levels of IL6 mRNA in 6 cases of MsPGN, 5 cases of non-MsPGN in renal biopsy and 4 cases of normal renal tissues were detected by reverse transcriptionpolymerase chain reaction. Results: The expression of IL6mRNA in kidney tissues of MsPGN and non-MsPGN patients were (x ± s) 0433 ± 0111 and 0271 ± 0023, respectively. MsPGN group IL 6 expression was significantly higher than non-MsPGN group (P <0 02). 4 cases of normal renal tissue in 3 cases without IL 6 mRNA expression, the remaining 1 cases the expression level was 0192. The moderate MsPGN and mild MsPGN expression levels were 0491 ± 0084 and 0317 ± 0018, the ratio of the two was 15: 1, MsPGN with focal segmental sclerosis of glomerulus and not The expression levels in patients with sclerotic lesions were 0516 ± 0083 and 0350 ± 0058, respectively, with the ratio of 147:1. In addition, the activity of IL-6 in urine of MsPGN children was determined by bioactivity assay. The activity of IL-6 in urine was positively correlated with the expression of IL-6 mRNA in renal tissues (r = 0.977, P <0.001) . Conclusion: IL 6 may be related to the pathogenesis of MsPGN, IL 6 may be involved in MsPGN mesangial proliferation and sclerosis, urinary IL 6 activity can reflect the local tissue expression of IL 6 mRNA
