目的:研究煎煮时间对附子水煎液总生物碱和6种酯型生物碱含量的影响。方法:采用酸碱滴定法测定总生物碱含量;采用HPLC,Agilent ZORBAX Extend-C18分析柱测定6种酯型生物碱含量。结果:随着煎煮时间的延长,不同炮制品中总生物碱的溶出率变化不明显;不同炮制品煎煮过程中酯型生物碱的变化规律不尽相同,其中乌头碱(AC)含量最低,水解最为彻底。结论:临床大剂量应用附子时,最大限度发挥其药理作用,恰如其分地掌握剂量与煎煮时间,是必不可少的。
Objective: To study the effects of decocting time on total alkaloids and six kinds of ester alkaloids in Aconiti decoction. Methods: The total alkaloids content was determined by acid-base titration. The content of six kinds of ester-type alkaloids was determined by HPLC and Agilent ZORBAX Extend-C18 column. Results: With the prolongation of boiling time, the dissolution rate of total alkaloids in different processed products did not change obviously. The changes of ester alkaloids in different processed products were different, and the contents of aconitine (AC) Lowest, the most complete hydrolysis. Conclusions: It is essential to apply large doses of aconite to maximize its pharmacological effects and to properly grasp the dosage and decoction time.
