目的:建立一种用反相高效液相色谱法测定中药材柴胡中柴胡皂苷d含量的方法,为柴胡的质量评价提供依据。考察六个不同产地柴胡中柴胡皂苷D的含量。方法:采用YMC pack ODS-A C18 150×4.6mm5μm色谱柱,流动相为甲醇-水(68:32),流速1.0ml/min,检测波长为 210nm,HPll00色谱仪,Agilent ChemStation工作站,外标法定量,以柴胡皂苷d含量为指标进行比较。结果:不同产地柴胡中的柴胡皂苷d含量差异悬殊,其中山西万荣所产柴胡中含量最高。结论:研究所建立的方法适合不同产地的柴胡中柴胡皂苷d的定量分析,并且简便可行,重现性好,结果准确可靠。
Objective: To establish a method for the determination of saikosaponin d content in Radix Bupleuri by reversed-phase high performance liquid chromatography, which will provide basis for quality evaluation of Radix Bupleuri. To investigate the content of saikosaponin D in Radix Bupleuri from different origins. METHODS: A YMC pack ODS-A C18 150×4.6mm 5μm column was used. The mobile phase was methanol-water (68:32), flow rate was 1.0ml/min, detection wavelength was 210nm, HPll00 chromatograph, Agilent ChemStation workstation. The standard method was used for quantification, and the content of saikosaponin d was used as an index for comparison. Results: The content of saikosaponin d in Bupleurum chinense from different producing areas was very different, among which the content of Bupleurum chinense produced by Wanrong was the highest. Conclusion: The method established in the institute is suitable for the quantitative analysis of saikosaponin d in Bupleurum chinense from different places, and it is simple and feasible, with good reproducibility and accurate and reliable results.
