研究了阴离子交换色谱中色谱柱温度对氨基酸保留的影响。实验采用氢氧化钠和醋酸钠作为淋洗液在Dionex AminoPac PA10阴离子交换柱上分离氨基酸,色谱柱温度为20~40℃。氨基酸的保留表现为吸热行为(随温度升高,保留增强)或放热行为(随温度升高,保留减弱)。温度对弱保留氨基酸的保留有明显影响。利用温度对弱保留氨基酸保留影响上的差异,通过改变柱温可以改善两种相近洗脱氨基酸之间的分离状况。温度对强保留氨基酸的保留无明显影响。弱保留氨基酸的范特霍夫曲线(lnk与1/T关系曲线)具有良好的线性关系。
The effect of column temperature on the retention of amino acids in anion exchange chromatography was investigated. The experiment used sodium hydroxide and sodium acetate as eluent to separate amino acids on Dionex AminoPac PA10 anion exchange column. The temperature of the column was 20 ~ 40 ℃. Amino acid retention shows endothermic behavior (with increasing temperature, increasing retention) or exothermic behavior (decreasing with increasing temperature). Temperature has a significant effect on the retention of weakly conserved amino acids. Using the effect of temperature on the retention of amino acids with weak retention, the separation between two closely eluted amino acids can be improved by changing the column temperature. No significant effect of temperature on the retention of strongly conserved amino acids. The van der Hoff curve of weakly conserved amino acids (lnk versus 1 / T) has a good linear relationship.
