为研究孕妇外周血中转铁蛋白受体阳性(TfR+)细胞中胎儿细胞的性质和来源,采用TfR的单克隆抗体(单抗)作为标记物,经免疫细胞化学检测,在35名8~20周妊娠孕妇外周血单个核细胞中检测到阳性细胞;同时在胎脐血中及胎盘滋养层细胞表面也检测到了相应的阳性细胞。在此基础上,对另外10名孕妇外周血细胞进行了多参数流式细胞术分析,发现了两个不见于正常成年人外周血的细胞亚群,即TfR+/中到高度FSC/低度SSC和TfR+/极高度FSC/高度SSC,并推测前者属幼稚红细胞性质,后者属滋养层细胞性质。采用荧光激活细胞分离技术对5例已证实怀男胎的孕妇外周血中TfR+细胞进行了分选及Y探针的荧光原位杂交鉴定,发现阳性细胞仅存在于TfR+细胞中。结果表明,孕妇外周血循环TfR+细胞中包含了胚胎幼稚红细胞和滋养层细胞。
In order to study the nature and origin of fetal cells in transferrin receptor positive (TfR +) cells from peripheral blood of pregnant women, TfR monoclonal antibody (mAb) was used as a marker and detected by immunocytochemistry in 35 8-20 weeks Positive cells were detected in the peripheral blood mononuclear cells of pregnant women. Corresponding positive cells were detected in the fetal umbilical cord blood and on the surface of placental trophoblast cells. On this basis, the other 10 pregnant women were analyzed by multi-parameter flow cytometry of peripheral blood cells and found two cell subsets not seen in peripheral blood of normal adults, namely TfR + / moderate to high FSC / low SSC and TfR + / very high level of FSC / height of SSC, and speculated that the former is an immature erythrocyte nature, which is a trophoblast cell nature. TfR + cells from 5 pregnant women with proven male fetus were identified by fluorescence activated cell sorting and fluorescent in situ hybridization with Y probe. The positive cells were found only in TfR + cells. The results show that pregnant women with peripheral blood TfR + cells contain embryonic naive erythrocytes and trophoblast cells.
