目的观察光照应激状态下SD大鼠睾丸组织中annexin5的表达变化。方法建立SD大鼠光照应激模型,24h持续光照,分为光照应激短期(2、3、4、7d)和光照应激长期(14、21、28d),分别取应激状态和相应对照组的大鼠睾丸组织,采用免疫组织化学和Western blotting技术分析annexin5免疫定位和蛋白相对表达。结果Western blotting结果显示:短期光照应激组(2、3、4、7d)的实验组大鼠睾丸annexin5表达与对照组相比无明显变化;在持续光照14d和21d后,实验组大鼠睾丸annexin5的相对表达量与对照组相比分别从3.09提高到4.16,从4.20提高到5.28(P<0.01,P<0.05);而持续光照28d后,实验组大鼠睾丸annexin5的相对表达量(4.87)与对照组(4.13)比较无明显差异(P>0.05)。免疫组织化学显示,annexin5在大鼠睾丸中的分布未见明显改变,但光照应激长期实验组(14、21、28d)annexin5免疫组化显色较深,说明annexin5表达升高,这与Westernblotting的结果基本一致。结论长期光照应激使大鼠睾丸annexin5的表达量明显增加。annexin5参与了应激过程,并可能参与了睾丸中某些重要生理功能的调节。
Objective To observe the changes of annexin5 expression in the testis of SD rats exposed to light. Methods The model of SD rats was established and exposed to light for 24 hours. Immunohistochemistry showed that there was no significant change in the distribution of annexin5 in rat testis, but the immunohistochemical staining of annexin5 in long-term light exposure group (14, 21, 28d) was darker, indicating that the annexin5 expression was increased, which was associated with Westernblotting The results are basically the same. Conclusion Long-term light stress can significantly increase the expression of annexin5 in rat testes. Annexin5 is involved in stress processes and may be involved in the regulation of certain important physiological functions in the testes.
